Forensic STR genotyping protocols often require long PCR cycling times which significantly slow down DNA sample processing. The goal of this study was to develop a fast protocol that produces complete and interpretable Identifiler profiles in 26 min. This was achieved by replacing AmpliTaq Gold with SpeedSTAR HS DNA polymerase, modifying the cycling parameters (shortened hold times at each temperature step), and adopting a two-step PCR approach.
